The PTCH1 c.4255C>T (p.Arg1419Trp) variant has been reported in the published literature in an individual with soft tissue and bone sarcoma (PMID: 37353797 (2023)). The frequency of this variant in the general population, 0.00012 (3/24948 chromosomes in African/African American subpopulation (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is higher than would generally be expected for pathogenic variants in this gene. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

The p.R1419W variant (also known as c.4255C>T), located in coding exon 23 of the PTCH1 gene, results from a C to T substitution at nucleotide position 4255. The arginine at codon 1419 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
