Inborn genetic diseases Uncertain:1
The p.N227K variant (also known as c.681T>A), located in coding exon 1 of the NHLRC1 gene, results from a T to A substitution at nucleotide position 681. The asparagine at codon 227 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species, and lysine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
NHLRC1-related disorder Benign:1
This variant is classified as likely benign based on ACMG/AMP sequence variant interpretation guidelines (Richards et al. 2015 PMID: 25741868, with internal and published modifications). -
