The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. A literature search was performed for the gene, cDNA change, and amino acid change (if applicable). No publications were found based on this search. This sequence change has been described in the gnomAD database with a frequency of 0.23% in the African American/African subpopulation (dbSNP rs141268133). The p.Arg365Ser change affects a poorly conserved amino acid residue located in a domain of the FANCE protein that is not known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Arg365Ser substitution. Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Arg365Ser change remains unknown at this time. -
