Hereditary cancer-predisposing syndrome Uncertain:2
The p.V429A variant (also known as c.1286T>C), located in coding exon 11 of the MRE11A gene, results from a T to C substitution at nucleotide position 1286. The valine at codon 429 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Ataxia-telangiectasia-like disorder Uncertain:1
This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 429 of the MRE11 protein (p.Val429Ala). This variant is present in population databases (rs141293060, gnomAD 0.09%). This missense change has been observed in individual(s) with an unspecified cancer (PMID: 28873162). ClinVar contains an entry for this variant (Variation ID: 142344). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
MRE11-related disorder Uncertain:1
The MRE11 c.1286T>C variant is predicted to result in the amino acid substitution p.Val429Ala. This variant was detected in one patient from a cohort of individuals with advanced cancer (eTable, Mandelker et al. 2017. PubMed ID: 28873162). This variant is reported in 0.084% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-94194142-A-G) and has been interpreted as uncertain in ClinVar (https://preview.ncbi.nlm.nih.gov/clinvar/variation/142344/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
