The p.I300V variant (also known as c.898A>G), located in coding exon 9 of the SBF2 gene, results from an A to G substitution at nucleotide position 898. The isoleucine at codon 300 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 4 Uncertain:1
This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 300 of the SBF2 protein (p.Ile300Val). This variant is present in population databases (rs141669540, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with SBF2-related conditions. ClinVar contains an entry for this variant (Variation ID: 216778).
