Mutations in this gene are associated with HMG-CoA synthase deficiency. This variant is present in population databases (rs142637231, gnomAD 0.005%). This premature translational stop signal has been observed in individual(s) with clinical features of HMGCS2-related conditions (PMID: 30283815, 32259399). ClinVar contains an entry for this variant (Variation ID: 280473). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
