This variant is present in population databases (rs143284092, gnomAD 0.2%), including at least one homozygous and/or hemizygous individual. This missense change has been observed in individual(s) with steroid-resistant nephrotic syndrome (PMID: 33749661). ClinVar contains an entry for this variant (Variation ID: 345989). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects LAMB2 function (PMID: 33749661).
