Genes affected
TMEM43 (HGNC:28472): (transmembrane protein 43) This gene belongs to the TMEM43 family. Defects in this gene are the cause of familial arrhythmogenic right ventricular dysplasia type 5 (ARVD5), also known as arrhythmogenic right ventricular cardiomyopathy type 5 (ARVC5). Arrhythmogenic right ventricular dysplasia is an inherited disorder, often involving both ventricles, and is characterized by ventricular tachycardia, heart failure, sudden cardiac death, and fibrofatty replacement of cardiomyocytes. This gene contains a response element for PPAR gamma (an adipogenic transcription factor), which may explain the fibrofatty replacement of the myocardium, a characteristic pathological finding in ARVC. Benign, criteria provided, single submitterclinical testingLaboratory for Molecular Medicine, Mass General Brigham Personalized MedicineSep 24, 2019The p.Leu369Phe variant in TMEM43 is classified as benign because it has been identified in 0.1% (27/24964) of African chromosomes by gnomAD (http://gnomad.broadinstitute.org). Additionally, 5 species (cape elephant shrew, chicken, soft shell turtle, spiny soft shell turtle, and nile tilapia) carry a phenylalanine (Phe) at this position at this position despite high nearby amino acid conservation, suggesting that this change may be tolerated. Computational prediction tools support that this variant does not impact the protein. ACMG/AMP Criteria applied: BA1, BP4. -
