This sequence change replaces cysteine, which is neutral and slightly polar, with phenylalanine, which is neutral and non-polar, at codon 365 of the LIG4 protein (p.Cys365Phe). This variant is present in population databases (rs144322912, gnomAD 0.08%). This variant has not been reported in the literature in individuals affected with LIG4-related conditions. ClinVar contains an entry for this variant (Variation ID: 533822). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant is not likely to affect RNA splicing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

DNA sequence analysis of the LIG4 gene demonstrated a sequence change, c.1094G>T, in exon 2 that results in an amino acid change, p.Cys365Phe.This sequence change has been described in the gnomAD database with a frequency of 0.08% in the East Asian subpopulation (dbSNP rs144322912). The p.Cys365Phe change affects a moderately conserved amino acid residue located in a domain of the LIG4 protein that is known to be functional. In-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL) provide contradictory results for the p.Cys365Phe substitution. This sequence change does not appear to have been previously described in individuals with LIG4-related disorders. Due to insufficient evidence and the lack of functional studies, the clinical significance of the p.Cys365Phe change remains unknown at this time. -
