The p.Asp1736Glu variant in DMD has not been previously reported in individuals with cardiomyopathy, but has been identified in 1/8394 African chromosomes by th e Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs14 4717862). Aspartic acid (Asp) at position 1736 is conserved in mammals but not i n evolutionarily distant species, and Xenopus tropicalis carries a glutamic acid (Glu) at this position, raising the possibility that this change may be tolerat ed. However, this information is not predictive enough to rule out pathogenicity . In summary, the clinical significance of the p.Asp1736Glu variant is uncertain . -
