The c.1298G>A (p.S433N) alteration is located in exon 11 (coding exon 10) of the DIS3L2 gene. This alteration results from a G to A substitution at nucleotide position 1298, causing the serine (S) at amino acid position 433 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. This sequence change replaces serine with asparagine at codon 433 of the DIS3L2 protein (p.Ser433Asn). The serine residue is highly conserved and there is a small physicochemical difference between serine and asparagine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a DIS3L2-related disease.
