The p.S812F variant (also known as c.2435C>T), located in coding exon 19 of the PLEKHG5 gene, results from a C to T substitution at nucleotide position 2435. The serine at codon 812 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces serine with phenylalanine at codon 812 of the PLEKHG5 protein (p.Ser812Phe). The serine residue is moderately conserved and there is a large physicochemical difference between serine and phenylalanine. This variant is present in population databases (rs145009237, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with PLEKHG5-related conditions.
