This alteration has been identified in individuals with a personal and/or family history of paragangliomas or pheochromocytomas (PGL/PCC), including tumors with a loss of fumarate hydratase staining in immunohistochemistry experiments (Ambry internal data; Fuchs TL et al. Am J Surg Pathol 2023 Jan;47(1):25-36; Zavoshi S et al. Urology 2023 Feb.). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the supporting evidence, this alteration is interpreted as pathogenic in association with PGL/PCC, however its association with other features of hereditary leiomyomatosis and renal cell carcinoma syndrome is unclear. - |
