Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00028 in 1611122 control chromosomes, including 3 homozygotes, predominantly at a frequency of 0.0037 within the African or African-American subpopulation in the gnomAD database, suggesting that the variant is a benign polymorphism. To our knowledge, no occurrence of c.5072C>T in individuals affected with ADGRV1-Related Disorders and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 227407). Based on the evidence outlined above, the variant was classified as likely benign. -
