Mutations in this gene cause primary ciliary dyskinesia; a disorder characterized by defects of the axoneme in motile cilia and sperm flagella. The homolog of this gene was first identified in the blue-green algae Chlamydomonas as encoding a radial spoke protein that formed a structural component of motile cilia and flagella. This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 15 of the RSPH3 protein (p.Leu15Arg). This variant is present in population databases (rs147256615, gnomAD 0.2%). This variant has not been reported in the literature in individuals affected with RSPH3-related conditions. ClinVar contains an entry for this variant (Variation ID: 572130). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign".
