The p.L1645F variant (also known as c.4933C>T), located in coding exon 29 of the SPG11 gene, results from a C to T substitution at nucleotide position 4933. The leucine at codon 1645 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Hereditary spastic paraplegia 11 Uncertain:1
This sequence change replaces leucine with phenylalanine at codon 1645 of the SPG11 protein (p.Leu1645Phe). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and phenylalanine. This variant is present in population databases (rs147735958, ExAC 0.09%). This variant has not been reported in the literature in individuals affected with SPG11-related conditions.
