Genes affected
LRRK2 (HGNC:18618): (leucine rich repeat kinase 2) This gene is a member of the leucine-rich repeat kinase family and encodes a protein with an ankryin repeat region, a leucine-rich repeat (LRR) domain, a kinase domain, a DFG-like motif, a RAS domain, a GTPase domain, a MLK-like domain, and a WD40 domain. The protein is present largely in the cytoplasm but also associates with the mitochondrial outer membrane. Mutations in this gene have been associated with Parkinson disease-8. Submissions by phenotype

Autosomal dominant Parkinson disease 8 Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingInvitaeJul 13, 2017In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with LRRK2-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces valine with methionine at codon 401 of the LRRK2 protein (p.Val401Met). The valine residue is weakly conserved and there is a small physicochemical difference between valine and methionine. -
Inborn genetic diseases Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsNov 17, 2022The p.V401M variant (also known as c.1201G>A), located in coding exon 11 of the LRRK2 gene, results from a G to A substitution at nucleotide position 1201. The valine at codon 401 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
