This alteration results from a T to C substitution at nucleotide position 1730, causing the leucine (L) at amino acid position 577 to be replaced by a proline (P). This allele was reported in one heterozygous individual in population-based cohorts in the Genome Aggregation Database (gnomAD). This has been reported as a compound heterozygous finding in multiple unrelated children with IGHMBP2-related neuropathy (Grohmann, 2003; Maystadt, 2004; Cassini, 2019). Another alteration at the same codon, c.1730T>G (p.L577R), has been detected as a compound heterozygous finding in a 2 month old Chinese male with spinal muscular atrophy with respiratory distress 1 (SMARD1) (Han, 2015). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis.
