Genes affected
IGHMBP2 (HGNC:5542): (immunoglobulin mu DNA binding protein 2) This gene encodes a helicase superfamily member that binds a specific DNA sequence from the immunoglobulin mu chain switch region. Mutations in this gene lead to spinal muscle atrophy with respiratory distress type 1. [provided by RefSeq, Jul 2008]

Submissions by phenotype

Inborn genetic diseases Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingAmbry GeneticsDec 11, 2023The c.1730T>C (p.L577P) alteration is located in coding exon 12 of the IGHMBP2 gene. This alteration results from a T to C substitution at nucleotide position 1730, causing the leucine (L) at amino acid position 577 to be replaced by a proline (P). This allele was reported in one heterozygous individual in population-based cohorts in the Genome Aggregation Database (gnomAD). This has been reported as a compound heterozygous finding in multiple unrelated children with IGHMBP2-related neuropathy (Grohmann, 2003; Maystadt, 2004; Cassini, 2019). Another alteration at the same codon, c.1730T>G (p.L577R), has been detected as a compound heterozygous finding in a 2 month old Chinese male with spinal muscular atrophy with respiratory distress 1 (SMARD1) (Han, 2015). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, this alteration is classified as pathogenic. -
Autosomal recessive distal spinal muscular atrophy 1;C4015349:Charcot-Marie-Tooth disease axonal type 2S Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpMar 09, 2023For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Leu577 amino acid residue in IGHMBP2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 25280635; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on IGHMBP2 protein function. ClinVar contains an entry for this variant (Variation ID: 522868). This missense change has been observed in individuals with Charcot-Marie-Tooth disease and/or spinal muscular atrophy with respiratory distress (PMID: 14681881, 15108294, 30598237, 31020813). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 577 of the IGHMBP2 protein (p.Leu577Pro). -

Uncertain significance, criteria provided, single submitterclinical testingUndiagnosed Diseases Network, NIHFeb 07, 2017The c.1730T>C variant has been observed in multiple, unrelated, affected individuals with Charcot Marie-Tooth disease 2 (Grohmann K, Varon R, Stolz P, et al. 2003). In addition, multiple lines of computational evidence support a deleterious effect of the p.Leu577Pro variant on the gene or gene product. This individual has been reported in PMID: 31020813. -
