The Arg339Gln variant in ESPN has not been previously reported in individuals wi th hearing loss, but has been identified in 0.01% (1/8600) of European American chromosomes and in 0.01% (1/4406) of African American chromosomes by the NHLBI E xome Sequencing Project (http://evs.gs.washington.edu/EVS/; dbSNP rs148390614). Computational prediction tools and conservation analysis do not provide strong s upport for or against an impact to the protein. In summary, the clinical signif icance of the Arg339Gln variant is uncertain. -

The c.1016G>A (p.R339Q) alteration is located in exon 6 (coding exon 6) of the ESPN gene. This alteration results from a G to A substitution at nucleotide position 1016, causing the arginine (R) at amino acid position 339 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
