The encoded protein can bind NF-kappa-B-inducing kinase and I-kappa-B kinases through separate domains and assemble them into an active kinase complex. This sequence change replaces threonine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 1051 of the ELP1 protein (p.Thr1051Ala). This variant is present in population databases (rs148548795, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with ELP1-related conditions. ClinVar contains an entry for this variant (Variation ID: 526202). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The alanine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The p.T1051A variant (also known as c.3151A>G), located in coding exon 27 of the IKBKAP gene, results from an A to G substitution at nucleotide position 3151. The threonine at codon 1051 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
