The gene product has been proposed to be an adapter or docking molecule. Mutations in this gene result in autosomal recessive Charcot-Marie-Tooth disease type 4C, a childhood-onset neurodegenerative disease characterized by demyelination of motor and sensory neurons. Variant summary: SH3TC2 c.689T>C (p.Val230Ala) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00055 in 1613722 control chromosomes, predominantly at a frequency of 0.0098 within the Ashkenazi Jewish subpopulation in the gnomAD database, including 1 homozygotes. The observed variant frequency within Ashkenazi Jewish control individuals in the gnomAD database (v4) exceeds the estimated maximal expected allele frequency for a pathogenic variant in SH3TC2 causing Charcot-Marie-Tooth disease type 4C phenotype. To our knowledge, no occurrence of c.689T>C in individuals affected with Charcot-Marie-Tooth disease type 4C and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 219388). Based on the evidence outlined above, the variant was classified as likely benign. -
