The c.3726_3730dupAGGCA variant, located in coding exon 19 of the BRIP1 gene, results from a duplication of AGGCA at nucleotide position 3726, causing a translational frameshift with a predicted alternate stop codon (p.M1244Kfs*12). This alteration occurs at the 3' terminus of BRIP1 gene, is not expected to trigger nonsense-mediated mRNAdecay and results in the elongation of the protein by 6 amino acids. This frameshift impacts the last 6 amino acids of the native protein. The exact functional effect of the altered amino acids is unknown. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This variant inserts 5 nucleotides in exon 20 of the BRIP1 gene, creating a frameshift in the last coding exon and addition of 6 new amino acids before introducing a stop codon. This mutant transcript is predicted to escape nonsense-mediated decay. Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been reported in 1 individual age 70 years or older without cancer by FLOSSIES (https://whi.color.com/variant/17-59760676--TGCCT). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD).
