Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 6.8e-06 in 1614000 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in COL5A2 causing Ehlers-Danlos syndrome, classic type, 2 (6.8e-06 vs 3.1e-05), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1463A>G in individuals affected with Ehlers-Danlos syndrome, classic type, 2 and no experimental evidence demonstrating its impact on protein function have been reported.
