Fibrillar collagen molecules are trimers that can be composed of one or more types of alpha chains. Type V collagen is found in tissues containing type I collagen and appears to regulate the assembly of heterotypic fibers composed of both type I and type V collagen. This gene product is closely related to type XI collagen and it is possible that the collagen chains of types V and XI constitute a single collagen type with tissue-specific chain combinations. Variant summary: COL5A2 c.1463A>G (p.His488Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 6.8e-06 in 1614000 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in COL5A2 causing Ehlers-Danlos syndrome, classic type, 2 (6.8e-06 vs 3.1e-05), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.1463A>G in individuals affected with Ehlers-Danlos syndrome, classic type, 2 and no experimental evidence demonstrating its impact on protein function have been reported.
