The c.1780A>G (p.S594G) alteration is located in exon 2 (coding exon 1) of the FLRT1 gene. This alteration results from a A to G substitution at nucleotide position 1780, causing the serine (S) at amino acid position 594 to be replaced by a glycine (G). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This variant has not been reported in the literature in individuals with FLRT1-related disease. This sequence change replaces serine with glycine at codon 594 of the FLRT1 protein (p.Ser594Gly). The serine residue is highly conserved and there is a small physicochemical difference between serine and glycine. This variant is present in population databases (rs150249733, ExAC 0.002%). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
