The p.K251R variant (also known as c.752A>G), located in coding exon 10 of the COL3A1 gene, results from an A to G substitution at nucleotide position 752. The lysine at codon 251 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved through mammals, but arginine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces lysine with arginine at codon 251 of the COL3A1 protein. Computational prediction tools indicate that this variant has a neutral impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with COL3A1-related disorders in the literature. This variant has been identified in 2/250228 chromosomes in the general population by the Genome Aggregation Database (gnomAD).
