The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

The threonine at codon 49 is replaced by serine, an amino acid with similar properties. This alteration was reported in one healthy control from an arrhythmogenic right ventricular dysplasia/cardiomyopathy (ARVD/C) genetic testing study, but clinical details were limited (Kapplinger JD et al. J. Am. Coll. Cardiol., 2011 Jun;57:2317-27). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
