MSH2 (HGNC:7325): (mutS homolog 2) This locus is frequently mutated in hereditary nonpolyposis colon cancer (HNPCC). When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. ESE finder predicts that this variant may affect ESE site of SRp40. However, these predictions are not confirmed by experimental studies. This variant was found in 1/56728 control chromosomes at a frequency of 0.0000176, which does not significantly exceed maximal expected frequency of a pathogenic allele (0.0005683). In addition, one reputable database classified this variant as VUS.
