Variant summary: SCN10A c.1858G>A (p.Val620Ile) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 7.5e-05 in 1592490 control chromosomes, predominantly at a frequency of 0.0011 within the African or African-American subpopulation in the gnomAD database (v4.1). The observed variant frequency within African or African-American control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for a pathogenic variant in SCN10A causing SCN10A-Related Disorders phenotype. c.1858G>A has been observed in individuals affected with Brugada syndrome without cosegregation information (e.g. Di Resta_2015, Tambi_2021). These reports do not provide unequivocal conclusions about association of the variant with SCN10A-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 26220970, 33797273). ClinVar contains an entry for this variant (Variation ID: 407745).
