The c.3524G>A (p.S1175N) alteration is located in exon 35 (coding exon 34) of the FANCD2 gene. This alteration results from a G to A substitution at nucleotide position 3524, causing the serine (S) at amino acid position 1175 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Fanconi anemia Uncertain:1
This sequence change replaces serine with asparagine at codon 1175 of the FANCD2 protein (p.Ser1175Asn). The serine residue is weakly conserved and there is a small physicochemical difference between serine and asparagine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with FANCD2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The asparagine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function.
