Genes affected
CAMTA1 (HGNC:18806): (calmodulin binding transcription activator 1) The protein encoded by this gene contains a CG1 DNA-binding domain, a transcription factor immunoglobulin domain, ankyrin repeats, and calmodulin-binding IQ motifs. The encoded protein is thought to be a transcription factor and may be a tumor suppressor. However, a translocation event is sometimes observed between this gene and the WWTR1 gene, with the resulting WWTR1-CAMTA1 oncoprotein leading to epithelioid hemangioendothelioma, a malignant vascular cancer. The c.766C>T (p.Q256*) alteration, located in exon 8 (coding exon 8) of the CAMTA1 gene, consists of a C to T substitution at nucleotide position 766. This changes the amino acid from a glutamine (Q) to a stop codon at amino acid position 256. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. Based on data from the Genome Aggregation Database (gnomAD), the CAMTA1 c.766C>T alteration was not observed, with coverage at this position. Based on the available evidence, this alteration is classified as pathogenic. -
