The encoded protein is one of the components of a calcium channel, composed of a tetramer of the ryanodine receptor proteins and a tetramer of FK506 binding protein 1B proteins, that supplies calcium to cardiac muscle. Mutations in this gene are associated with stress-induced polymorphic ventricular tachycardia and arrhythmogenic right ventricular dysplasia. [provided by RefSeq, Jul 2008]

Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has been reported in an individual affected with catecholaminergic polymorphic ventricular tachycardia (CPVT) (PMID: 15175054). This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with valine at codon 2534 of the RYR2 protein (p.Leu2534Val). The leucine residue is highly conserved and there is a small physicochemical difference between leucine and valine. -
