The p.A157S variant (also known as c.469G>T), located in coding exon 10 of the TNNT2 gene, results from a G to T substitution at nucleotide position 469. The alanine at codon 157 is replaced by serine, an amino acid with similar properties. This alteration was reported in an individual with hypertrophic cardiomyopathy (Millat G et al. Eur J Med Genet Jul;53:261-7). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces alanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 157 of the TNNT2 protein (p.Ala157Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 20624503). ClinVar contains an entry for this variant (Variation ID: 519034). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TNNT2 protein function with a positive predictive value of 95%.
