When cloned, it was discovered to be a human homolog of the E. coli mismatch repair gene mutS, consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. This sequence change replaces asparagine with serine at codon 311 of the MSH2 protein (p.Asn311Ser). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and serine. -
