Uncertain significance, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Oct 27, 2016 | The p.Pro478Leu variant in SOS1 has been reported in 2 individuals with clinical features of Noonan syndrome, one of whom also carried an additional SOS1 varian t (p.Gln477His) in cis and both variants were de novo (Zenker 2007, Lepri 2011). This variant was absent from large population studies. An additional amino acid change at this position (p.Pro478Arg) has been reported to occur de novo in two probands, suggesting that a change at this position may not be tolerated (Zenke r 2007, Lepri 2011). Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein. In summary, while there is some suspicion for a pathogenic role, the clinical significance of the p.Pro478Leu variant is uncertain. - |
