This variant is predicted to cause a frameshift, which alters the protein?s ami no acid sequence beginning at position 392 and leads to a premature termination codon 18 amino acids downstream. This alteration is then predicted to lead to a truncated or absent protein. Frameshift and other loss of function variants have been reported in association with EDS in the HGMD database (Stenson, 2017). In summary, although additional studies are required to fully establish its clinica l significance, the p.Pro392fs variant is likely pathogenic. - |
