Bethlem myopathy 1A Uncertain:1
This sequence change replaces glycine with aspartic acid at codon 1722 of the COL6A3 protein (p.Gly1722Asp). The glycine residue is weakly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a COL6A3-related disease. This variant occurs outside of the conserved triple-helical domain of the type 6 alpha-3 collagen protein where amino acid substitutions are rarely pathogenic.  However, substitution of a nearby amino acid (p.Leu1726Arg) was found in an individual with Bethlem myopathy and the variant was shown to have arisen de novo in the patient (see Patient BM2 in PMID: 17886299). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. -
