The properties of the channel formed by the encoded transmembrane protein can be altered by interaction with different beta subunits. This variant has not been reported in the literature in individuals with SCN10A-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces methionine with isoleucine at codon 684 of the SCN10A protein (p.Met684Ile). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and isoleucine. -
