Colorectal cancer, hereditary nonpolyposis, type 2 Pathogenic:1
This variant is considered pathogenic. This variant creates a frameshift predicted to result in premature protein truncation. -
not provided Pathogenic:1
This deletion of one nucleotide in MLH1 is denoted c.940delG at the cDNA level and p.Val314PhefsX53 (V314FfsX53) at the protein level. The normal sequence, with the base that is deleted in brackets, is TGAA[delG]TTCA. The deletion causes a frameshift, which changes a Valine to a Phenylalanine at codon 314 and creates a premature stop codon at position 53 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. We consider this variant to be pathogenic. -
Hereditary nonpolyposis colorectal neoplasms Pathogenic:1
This sequence change deletes 1 nucleotide from exon 11 of the MLH1 mRNA (c.940delG), causing a frameshift at codon 314. This creates a premature translational stop signal (p.Val314Phefs*53) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in MLH1 are known to be pathogenic (PMID: 15713769, 24362816). For these reasons, this variant has been classified as Pathogenic. -
Hereditary cancer-predisposing syndrome Pathogenic:1
The c.940delG pathogenic mutation, located in coding exon 11 of the MLH1 gene, results from a deletion of one nucleotide at nucleotide position 940, causing a translational frameshift with a predicted alternate stop codon. Since frameshifts are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
