The encoded protein functions as a transcriptional repressor during embryogenesis through interactions with components of the core transcription complex and other homeoproteins. It may also have roles in limb-pattern formation, craniofacial development, particularly odontogenesis, and tumor growth inhibition. Mutations in this gene, which was once known as homeobox 7, have been associated with nonsyndromic cleft lip with or without cleft palate 5, Witkop syndrome, Wolf-Hirschom syndrome, and autosomoal dominant hypodontia. Therefore, it has been classified as Pathogenic. This variant has not been reported in the literature in individuals with a MSX1-related disease. However, a different truncation downstream of this variant (p.Asn222Lysfs*118) has been determined to be pathogenic (PMID: 23991204). This suggests that deletion of this region of the MSX1 protein is causative of disease. This sequence change results in a premature translational stop signal in the last exon of the MSX1 mRNA at codon 221 (p.Gln221*). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 83 amino acids of the MSX1 protein. -
