Variant summary: IDUA c.1154C>G (p.Pro385Arg) results in a non-conservative amino acid change located in the Glycosyl hydrolases family 39, N-terminal catalytic domain (IPR049166) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 158834 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1154C>G has been reported in the literature in the compound heterozygous state in at least one individual affected with Mucopolysaccharidosis Type 1 and in one individual with an abnormal newborn screen who had normal glycosaminoglycans (Bertola_2011, Fillman_2023). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 21394825, 37516270). ClinVar contains an entry for this variant (Variation ID: 556024). Based on the evidence outlined above, the variant was classified as uncertain significance. -
Inborn genetic diseases Uncertain:1
The c.1154C>G (p.P385R) alteration is located in exon 8 (coding exon 8) of the IDUA gene. This alteration results from a C to G substitution at nucleotide position 1154, causing the proline (P) at amino acid position 385 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
