The c.506_510delTAGAT pathogenic mutation, located in coding exon 4 of the APC gene, results from a deletion of 5 nucleotides between nucleotide positions 506 and 510, causing a translational frameshift with a predicted alternate stop codon. While this exact alteration has not been reported in the literature, a different alteration, c.518delC, resulting in the same stop codon, has been detected in an individual affected with familial adenomatous polyposis (FAP) (Dobbie et al Eur. J. Cancer. 1994; 30A(11):1709-13). In addition to the clinical data presented in the literature, since frameshifts are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
