The p.W593* pathogenic mutation (also known as c.1779G>A), located in coding exon 14 of the APC gene, results from a G to A substitution at nucleotide position 1779. This changes the amino acid from a tryptophan to a stop codon within coding exon 14. This alteration was identified in 1/1164 unrelated German index patients with a clinical diagnosis of FAP or AFAP (Friedl W et al. Hered Cancer Clin Pract, 2005 Sep;3:95-114). This alteration was also detected in 1/150 FAP patients (Nagase H et al. Hum. Mutat., 1992;1:467-73). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
