The gene product has been proposed to be an adapter or docking molecule. Mutations in this gene result in autosomal recessive Charcot-Marie-Tooth disease type 4C, a childhood-onset neurodegenerative disease characterized by demyelination of motor and sensory neurons. This variant has not been reported in the literature in individuals with SH3TC2-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with methionine at codon 1033 of the SH3TC2 protein (p.Ile1033Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. -
