Variant summary: CFTR c.274-3T>C, also reported as c.406-3T>C and IVS3 406-3T>C, alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250472 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.274-3T>C has been observed in the presumed compound heterozygous state in at least 1 individual(s) affected with Cystic Fibrosis (Wong_2022, Ocak_2014) and also in the heterozygous state in individuals with clinical features of CFTR-related conditions (Alibakhshki_2008, Kilinc_2022) without strong evidence for causality. These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 17662673, 26437683, 38153325, 35072004, 25536748, 38055834, 34525262, 12439892, 16835862, 25525159).
