This sequence change replaces leucine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 842 of the KCNH2 protein (p.Leu842Pro). The p.L842P variant (also known as c.2525T>C), located in coding exon 10 of the KCNH2 gene, results from a T to C substitution at nucleotide position 2525. The leucine at codon 842 is replaced by proline, an amino acid with similar properties, and is located in the cAMP binding domain. Internal structural analysis indicates this alteration may disrupt the cAMP ligand binding domain (Wang W. Cell. 2017 Apr;169(3):422-430.e10). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
