This variant has been reported in the literature in multiple individuals with multiple osteochondromas (PMID: 17301954, 19810120, 24532482). This variant is not present in population databases (ExAC no frequency). This sequence change replaces arginine with glycine at codon 346 of the EXT1 protein (p.Arg346Gly). The arginine residue is highly conserved and there is a moderate physicochemical difference between arginine and glycine. -
