Primary ciliary dyskinesia Uncertain:2
This variant is not present in population databases (ExAC no frequency). This variant has been reported in an individual affected with primary ciliary dyskinesia (PMID: 21143860). In addition, this variant occurs with a second rare variant (p.Phe556Ser) in DNAI1 in an individual with primary ciliary dyskinesia (Invitae). While it is unknown if these two variants are on the same or opposite chromosomes, this observation suggests the c.1538T>C substitution may contribute to the cause of disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may alter RNA splicing, but this prediction has not been confirmed by published transcriptional studies. In summary, this variant is a rare missense change with uncertain impact on protein function. While it is absent from the population and reported in affected individuals, the available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces leucine with proline at codon 513 of the DNAI1 protein (p.Leu513Pro). The leucine residue is moderately conserved and there is a moderate physicochemical difference between leucine and proline. -
