The heterozygous c.2548-1G>A variant was identified by our study in one individual with congenital microcephaly, retinitis pigmentosa, and developmental delay. Trio exome analysis showed this variant to be de novo. This variant was absent from large population studies. The c.2548-1G>A variant occurs in the invariant region (+/- 1/2) of the splice consensus sequence and is predicted to cause altered splicing leading to an abnormal or absent protein. Loss of function of the KIF11 gene is an established disease mechanism in autosomal dominant microcephaly, and this is likely a loss of function variant. In summary, this variant is pathogenic. -
