The c.3720_3721delGT pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 3720 to 3721, causing a translational frameshift with a predicted alternate stop codon (p.F1241*). This variant was identified in two individuals from one family in a Norwegian HBOC cohort (Heramb C et al. Hered Cancer Clin Pract. 2018 Jan;16:3). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
