Variant summary: BRCA2 c.6373_6374delinsG (p.Thr2125AlafsX12) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 237640 control chromosomes (gnomAD). This variant has been reported in a patient affected with breast and/or ovarian cancer (UMD-BRCA2 database), in addition, overlapping truncating variants (e.g. c.6373delA, c.6373dupA, c.6373_6375delACCinsG) were also reported in several affected individuals (HGMD). To our knowledge, no experimental evidence demonstrating its impact on protein function have been reported. Three other submitters, including an expert panel (ENIGMA), have provided clinical-significance assessments for this variant in ClinVar after 2014, and classified the variant as pathogenic (2x; including the expert panel) or likely pathogenic (1x). Based on the evidence outlined above, the variant was classified as pathogenic. -
This sequence change creates a premature translational stop signal (p.Thr2125Alafs*12) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 438996). For these reasons, this variant has been classified as Pathogenic. -

The c.6373_6374delACinsG pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from the deletion of two nucleotides and insertion of one nucleotide causing a translational frameshift with a predicted alternate stop codon (p.T2125Afs*12). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
